Palmitic Acid Lipotoxicity in Microglia Cells Is Ameliorated by Unsaturated Fatty Acids

Obesity and metabolic syndrome are associated with cognitive decline dementia. Palmitic acid (PA) is increased in the cerebrospinal fluid of obese patients impairment. This study was therefore designed to examine fatty (FA) lipotoxicity BV2 microglia cells. We found that PA induced time- dose-dependent decrease cell viability increase death without affecting cycle profile did not depend on surface free receptors but rather FA uptake. Treatment sulfosuccinimidyl oleate (SSO), an irreversible inhibitor translocase CD36, significantly inhibited uptake BSA- PA-treated cells blocked PA-induced viability. Inhibition ER stress or treatment N-acetylcysteine able rescue lipotoxicity. Our also showed unsaturated acids (UFAs), such as linoleic (LA), oleic (OA), ?-linolenic (ALA), docosahexaenoic (DHA), were lipotoxic instead protected against Co-treatment LA, OA, DHA All UFAs tested incorporation FAs into amount neutral lipids, while affect lipids compared BSA control.
